Ornithine Decarboxylase Resources
This page contains information about some of the reagents that we use in our studies of Ornithine Decarboxylase regulation, and a few links to other resources of interest to ODC and polyamine researchers.
ODC reporter plasmids
These plasmids were described in Kumar et al., 1995 and Mar et al., 1995. They are based on the luciferase reporter plasmid, pGL2-basic, available from Promega. This plasmid was modified to eliminate a cryptic AP-1 site in the SV40 sequences 3' of the luciferase sequences. This resulted in creation of an additional EcoRI site. The "founder" of this series is pODClux1m, which contains rat ODC sequences from -1156 to +13, relative to the transcription start site. (The blue cross-hatched region is the luciferase gene; the magenta color represents ODC promoter sequences.) Here is the complete sequence. (Either copy and paste from your browser window, or control-click to download the text file to your hard drive). The ODC sequences begin at position 38 in this plasmid.
Nested deletions derived from pODClux1m include pODClux2m (containing ODC sequences -412 to +13) and pODClux2 Em (ODC sequences -92 to +13). We have a number of additional deletions and site-directed mutations of the ODC promoter regions. For details, send an e-mail request to the address at the bottom of this page.
